Nidirana okinavana (Boettger, 1895) is a small-sized ranid species belonging to the East Asian genus Nidirana Dubois, 1992. Previous studies have indicated that this species was exclusively distributed on Ishigaki and Iriomote islands in the southern Ryukyus, as well as two extremely small wetland habitats in central Taiwan. Such a restricted distribution makes it one of the most endangered frog species in both Taiwan and Japan. By using molecular, morphological, and acoustic analyses, our study reveals significant divergence between the Taiwanese and Japanese clades, supporting the recognition of the Taiwanese clade as a distinct species, described herein as Nidirana shyhhuangi sp. nov. Compared to Nidirana okinavana sensu stricto from the southern Ryukyus, the Nidirana shyhhuangi sp. nov. is characterized by a significantly smaller and non-overlapping body size, relatively longer forelimbs and hindlimbs, smaller internostril and interorbital distances, with a higher number of cross bands on thigh and shank. Acoustic analyses reveal that the Nidirana shyhhuangi sp. nov. produces calls with a rapid tempo and higher pulse number, with a higher dominant frequency compared to the Japanese clade. Due to the extremely limited distribution of this species to two small sites on Taiwan, and continuing decline in quality of its habitat, we propose that it should be classified as Critically Endangered (CR) under the IUCN criteria. Immediate and comprehensive in situ and ex situ conservation actions are necessary to ensure the sustainable viability of the population.

<p>The Japanese giant salamander, <i>Andrias japonicus</i>, is one of the largest extant amphibians. It is endemic to Japan and has been designated as a special natural monument by the Japanese government. The genus <i>Andrias</i> is also protected by the Convention on International Trade in Endangered Species of Wild Fauna and Flora. The Japanese giant salamander and one of the congeners, the Chinese giant salamander, <i>A. davidianus</i>, were recently found to hybridize in several areas in Japan, due to human-mediated introduction of <i>A. davidianus</i>. In this study, we found major differences in head and skull shapes between the Japanese and Chinese giant salamanders, using geometric morphometrics. The hybrids showed intermediate morphology between the two species, but also possessed a wider mouth, at the skeletal level, than either of the parent species. This unique characteristic of hybrids is suggested as one of the reasons for the current dominance of the hybrids in the introduced areas and may be an example of heterosis in urodeles.</p>

The chytrid fungus Batrachochytrium salamandrivorans [Bsal] is causing declines in the amphibian populations. After a decade of mapping the pathogen in Europe, where it is causing dramatic outbreaks, and North America, where its arrival would affect to the salamander’s biodiversity hotspot, little is known about its current status in Asia, from presumably is native. Japan has several species considered as potential carriers, but no regulation is implemented against Bsal spreading. Previous Bsal known presence detected various cases on the Okinawa Island, southwestern Japan. Previous studies on its sister species, B. dendrobatidis presented a high genomic variation in this area and particularly on Cynops ensicauda. Here, we have done the largest monitoring to date in Japan on the Cynops genus, focusing on Okinawa Island and updating its distribution and providing more information to unravel the still unknown origin of Bsal. Interestingly, we have provided revealing facts about different detectability depending on the used molecular techniques and changes in its Japanese distribution. All in all, the Bsal presence in Japan, together with its low variability in the sequenced amplicons, and the lack of apparent mortalities, may indicate that this part of Asia has a high diversity of chytrids.

Abstract The modern and Late Pleistocene terrestrial fauna of Miyako Jima and adjacent islands (the Miyako Islands) in the southern Ryukyu Islands, southwestern Japan, includes some endemic taxa or genetically unique populations that exclusively have closest allies in the more isolated Okinawa Jima and adjacent islands (the Okinawa Islands) than in the Yaeyama Islands, which are located southwest of the Miyako Islands with much narrower intervening straits. Those taxa or populations include representatives of lineages that have physiologically highly limited ability for over-sea dispersal and the Miyako Islands are currently separated from the Okinawa Islands by at least 300 km of open water; therefore, the formation of this phylogeographical pattern is perplexing. In this study, we review the late Cenozoic geology of the Miyako Islands, southern Okinawa Jima, the Okinawa–Miyako submarine plateau (OMSP; a plateau located between Okinawa Jima and Miyako Jima), and the Kerama gap, which is a depression between the OMSP and Okinawa Jima. We then consider the origin of the modern and Late Pleistocene terrestrial animals, including a number of non-volant vertebrates on the Miyako Islands. Finally, we propose a new hypothesis (the OMSP hypothesis) to explain the enigmatic composition of modern and Late Pleistocene terrestrial vertebrate fauna of the islands. Southern Okinawa Jima was uplifted and emerged after ca. 2 Ma and was temporarily connected to the OMSP, which is likely to have emerged earlier than southern Okinawa Jima, to form a large island extending from Okinawa Jima to the Miyako Islands with a NE–SW direction of ~ 400 km. Subsequently, Okinawa Jima became separated from the OMSP when the Ryukyu Group—which is composed of Quaternary reef and associated fore-reef and shelf deposits—began to accumulate around the island at 1.7–1.4 Ma. During the interval from 2.0 to 1.7–1.4 Ma, numerous terrestrial animals, including flightless vertebrates, extended their distribution to the OMSP. Although the Miyako Islands repeatedly underwent complete submergence during deposition of the main part of the Ryukyu Group (1.25–0.4 Ma), they were uplifted and emerged to become a land area after ca. 0.4 Ma. In contrast, the OMSP subsided after ca. 0.4 Ma and was almost completely submerged after 0.27 Ma. During ca. 0.4–0.27 Ma, terrestrial animals migrated from the OMSP to the Miyako Islands.

<p>The Anderson’s crocodile newt, <i>Echinotriton andersoni</i>, is an endangered species distributed in the islands of Central Ryukyus, Japan. This species is well known to show high intraspecific genetic variation among islands, but its morphological variation has not been explored. We examined the skull variation in this species including enigmatic specimens collected from Taiwan by using geometric morphometrics, and found sexual differences and geographic variation mainly between the Okinawa group (Okinawajima Island) and the Amami Group (Amamioshima and Tokunoshima Islands). We further found close resemblance between the Taiwanese specimens and the female specimens from Amamioshima Island.</p>

<p>The family Rhacophoridae, including <i>Buergeria japonica</i>, shows a great diversity of reproductive patterns, but the knowledge of developmental processes is still limited. The genus <i>Buergeria</i> is a sister group to all other rhacophorids and shows a conservative, probably primitive, reproductive mode for this family. Thus, it is valuable to clarify the developmental process in this genus for understanding the evolution and diversification history of reproductive modes and developmental processes across the broader family members. In this study, we describe the normal development of <i>B. japonica</i> by rearing eggs and larvae under ambient temperatures of 27±1°C. The developmental speed of <i>B. japonica</i> from fertilization to gill elongation (stage 20), corresponding to their hatching period, was faster than most of other anuran species, when comparison was made using relative age, which is independent of temperature. The rapid embryonic development may be advantageous in their highly fluctuating breeding environment.</p>

<p>The diet of the nonnative American Bullfrog <i>Lithobates catesbeianus</i> occurring in Ogimi Village of Okinawajima, Ryukyu Archipelago, is investigated. Seventy two of 89 frogs (nine adults, five subadults, and 58 juveniles) captured had food items in its stomach. We identified a total of 64 taxa from 253 food items. Our analyses show that (1) the diet consists mainly of terrestrial prey, (2) mollusks and vertebrates are the essential prey groups for adults and subadults, and (3) odonates, mollusks, and arachnids are the main prey for juveniles. Such dietary habits are quite different from those of several conspecific nonnative populations in mainland Japan and other regions. Part of this is most likely related to the absence of the frog's favorite prey, nonnative American crayfish <i>Procambarus clarkii</i>, in the study area. We also confirm the predation of a poisonous newt <i>Cynops ensicauda popei</i> (Salamandridae) and several aquatic insects by this frog and present some implications for these results.</p>

<p>The abundant clouded salamander from western Japan, <i>Hynobius nebulosus</i>, has long been considered a single widespread species, although some authors have suggested the inclusion of several cryptic species. This led to a molecular and morphological analysis of populations from all parts of the known range. Phylogenetic relationships were inferred from complete sequences of the mitochondrial cytochrome b gene, and nuclear genome differentiations were estimated by multiplexed inter simple sequence repeat genotyping by sequencing (MIG-seq). The results suggest that <i>H. nebulosus</i> hitherto recognized consists of at least nine species. We applied existing names to two of them, <i>H. nebulosus</i> (Temminck et Schlegel) and <i>H. vandenburghi</i> Dunn, and described seven others as new species.</p>

<p>The Japanese giant salamander, <i>Andrias japonicus</i>, is known to exhibit very low genetic diversity, but the number of individuals surveyed in a population is limited by now. We investigated partial sequences (673 bp) of mitochondrial cytochrome b gene in 180 specimens of a population from the Inuyama Head Works on the Kiso River, Central Japan, over nine years so as to clarify the degree of genetic diversity. The result again confirmed a tendency of lack of diversity; only one individual differed from the remaining 179 with an uncorrected p-distance of only 1.5%. The individual had the sequence identical with that reported for an individual from San'in District of Western Japan, far from the Kiso River, and is thought to have been introduced artificially. The healthy condition of the salamander population in spite of highly reduced genetic diversities might be due to possible decrease of inbreeding depression, resulting from the past purging effect of ancestral inbreeding wherein deleterious recessive alleles were eliminated from the gene pool.</p>

Spontaneous oral chytridiomycosis in wild bullfrog tadpoles in Japan.

<i>Batrachochytrium dendrobatidis</i> (Bd) infects Anuran larvae (tadpole) mouthparts and causes oral chytridiomycosis, which can be diagnosed in tadpoles by detecting mouthparts deformities. However, oral chytridiomycosis may or may not be observable, depending on species, tadpole stage and season, and has never been reported in Japan. We aimed to observe oral chytridiomycosis characteristics in bullfrog (<i>Lithobates catesbeiana</i>) tadpoles, determine associated pathologic features and investigate the usability of bullfrog tadpoles in Japanese Bd field surveys. Wild-captured bullfrog tadpole mouthparts were examined macroscopically, histopathologically and by molecular biological examination. Macroscopic lesions were observed in 21 of 59 tadpole mouthparts. Lesions were most frequently located in the lower jaw sheaths and were mainly recognized by partial depigmentation (11 tadpoles; some were completely depigmented) and thinning of the pigmented layer (10 tadpoles). Partial defects of the tips and blunt cutting edges of the jaw sheaths were observed with severe jaw sheath depigmentation. Whitened tooth rows were observed in 7 tadpoles. Histologically, the stratified epithelium (pigmented epithelium) showed partial or diffuse hypopigmentation or pigment loss. Irregular stratified epithelium thickening with hyperkeratosis or parakeratosis was observed in the jaw sheaths. Bd infection was confirmed in 20 of 21 tadpoles presenting jaw sheath deformities, by histopathological examination and/or nested polymerase chain reaction. Depigmentation and thinning of the pigmented layers of jaw sheaths were associated with Bd infection. Thus, diagnosis of Bd infection by macroscopic observation of bullfrog tadpole mouthparts is feasible. This is the first report of oral chytridiomycosis in wild bullfrog tadpoles in Japan.

Using mitochondrial DNA sequences, we surveyed genetic features of 12 individuals of Cynops pyrrhogaster introduced into Hachijojima Island, in order to estimate their possible origin in the main islands of Japan. The phylogenetic analysis revealed that the 12 examined newts of the introduced population display no genetic diversity and share characteristics with the conspecific population of Shikoku and thus belong to the WESTERN Clade proposed by earlier studies. Our results suggest that the Hachijojima population, having experienced a drastic founder effect, stems from very few individuals from the Shikoku District approximately 650 km to the west.

Occurrence and Evolutionary History of Two <i>Cynops pyrrhogaster</i> Lineages on the Izu Peninsula

We investigated phylogenetic positions of Cynops pyrrhogaster from nine localities in the central and southern parts of the Izu Peninsula using the mitochondrial cyt b gene. We revealed that the central and the southern populations are phylogenetically remote. The central Izu lineage belongs to the CENTRAL clade occurring from Chubu through Kinki to Chugoku districts, whereas the southern Izu populations form a lineage sister to the NORTHERN clade, which is distributed in Tohoku and Kanto districts. Genetic differentiation between the southern Izu lineage and the NORTHERN clade is relatively large with the uncorrected p-distance of 3. 4%, which suggests their divergence at 3. 31 MYA. This estimation indicates their genetic differentiation prior to 1. 0 MYA, when the Izu Peninsula was formed through collision of a paleo-oceanic island with Honshu. These results indicate that the ancestor of the southern Izu lineage diverged from the NORTHERN clade somewhere in northern Honshu and then invaded the Izu Peninsula newly formed by collision and settled there. The central Izu lineage thereafter also invaded the peninsula, confining the range of the preceding southern Izu lineage to its current range.
