【総説-受賞論文-】　植物細胞壁多糖の生合成ならびに構造解析に関する研究

　概要を見る

植物の細胞壁多糖構造を解明するには,細胞壁を構成する多糖を緻密に分離し,それぞれの多糖について解析を行う必要がある。しかし,キシログルカンのような数種の糖から構成されるヘテロ多糖の構造は極めて複雑であり,構造解析には多くの時間とサンプル量を要する。そこで,キシログルカン特異的な酵素であるキシログルカナーゼおよびイソプリメベロース生成酵素を用いた新たな構造解析方法を開発し,迅速かつ微量サンプルでのキシログルカン構造解析を可能とした。一方,細胞壁多糖の生合成メカニズムの解明は,細胞壁構造を理解するうえで重要なアプローチであるが,その生合成メカニズムは未だ不明な点が多い。細胞壁多糖の多くはピラノース型の糖から構成されているのに対し,フラノース型の糖のみで構成されているアラビナンの生合成メカニズムは,植物細胞壁多糖の中でも最も興味深いものである。筆者はin vitroにおけるアラビナンの生合成に世界で初めて成功した。ついで,アラビナンの生合成基質となるUDP-アラビノフラノースを生成する酵素(UDP-アラビノピラノースムターゼ)の精製・単離に成功し,本酵素が植物に存在することを証明した。
