Development of the Male-sterile Line of Eggplant Utilizing the Cytoplasm of Solanum aethiopicum L. Aculeatum Group

In order to develop a new male-sterile line of eggplant (Solanum melongena L.), the F-1 rootstock 'Assist' (Solanum aethiopicum Aculeatum L. Group x S. melongena 'DMP') was continuously backcrossed to S. melongena 'Uttara' using 'Uttara' as a recurrent pollen parent up to BC4 generation. All examined F-1 'Assist' produced pollen grains while BC1 plants segregated into male fertile and sterile plants. The male-sterile plant did not produce any pollen grains. BC2, BC3, and BC4 progenies obtained from male fertile backcross progenies were segregated into male fertile and sterile plants and each segregation ratio fitted 3:1. BC3 and BC4 progenies obtained from male-sterile backcross progenies were fixed to male-sterility without segregation. Selfed progenies of the male fertile BC2 progeny were segregated into male fertile and sterile plants and the segregation ratio fitted 15:1. Therefore, the pollen non-formation characteristic is assumed to be a form of cytoplasmic male-sterility (CMS), induced by incompatibility between the cytoplasm of S. aethiopicum Aculeatum Group and the nucleus of S. melongena. Further, these segregation results indicated that two independent dominant fertility restorer (Rf) genes control the fertility restoration of this CMS. High values of fruit set, number of seeds per fruit, and the seed germination rate found in BC4 progenies indicate that the cytoplasm of S. aethiopicum Aculeatum Group has no notable negative effect on seed fertility of S. melongena. A new male-sterile line of eggplant could be developed by utilizing the cytoplasm of S. aethiopicum Aculeatum Group.

In order, to develop a male-sterile eggplant (Solanum melongena L.), cytoplasm. substitution lines of eggplant were produced by continuous backcrossing using S. kurzii Brace & Prain as cytoplasm donor and eggplant as nucleus donor. Analyses of chloroplast DNA and mitochondrial DNA confirmed that all backcross progenies had the cytoplasm from S. kurzii. Anthers in some BC(1) and BC(2) plants, and in most BC(3) plants were. of the indehiscent-type (i.e., pollen non-release type). Pollen, fertility remained low in all backcross progenies. The number of seeds per fruit in the backcross progenies increased in successive generations. Functional male-sterility of. the anther- indehiscent-type Was shown, to be attributed to. incompatibility between the cytoplasm of S. kurzii and nuclear genes of S. melongena. The present. male-sterile lines are potentially valuable to reduce the cost of developing hybrid cultivars' and to produce. seedless eggplant fruit.

Inter-simple sequence repeat (ISSR) analysis was performed in eight cultivars of eggplant (Solanum melongena) and 12 accessions in eight related Solanum species to evaluate the applicability of this analysis for assessing the phylogenetic relationships and identifying cultivars. A total of 552 polymorphic amplified bands were obtained from 34 of the 100 primers tested, and the percentage of polymorphisms was 99.1%. Cluster analysis based on the ISSR markers classified the Solanum species into seven groups: (i) S. melongena; (ii) S. aethiopicum and S. anguivi; (iii) S. incanum; (iv) S. violaceum and S. kurzii; (v) S. macrocarpon; (vi) S. virginianum and (vii) S. torvum. Combining the ISSR markers obtained by a few of the 34 primers was enough for distinguishing of the eight cultivars of eggplant. This ISSR analysis was demonstrated to be available for the phylogenetic study and the cultivar identification. (C) 2008 Elsevier B.V. All rights reserved.
